Mast cells (MCs) are multifunctional, granulated, stationary cells that reside in almost all vascularized tissues. Upon activation, MCs degranulate and release a large array of pro-inflammatory and vasoactive mediators and signaling molecules including histamine, prostaglandins, and leukotrienes. Inappropriate release of MC mediators causes so-called mast cell mediator-related symptoms and plays an essential role in many human diseases.


MCs are well-known for their role as the effector cells of immediate-type hypersensitivity reactions; however, they also play a crucial role in the pathogenesis of MC disorders, such as mastocytosis. MC activation may occur at the local level, such as in urticaria, asthma or systemically with clinical signs and symptoms of anaphylaxis. Anaphylaxis concurrently affects multiple organ systems and presents with a broad array of symptoms and signs. There is strong evidence of an association between severe anaphylaxis, especially hymenoptera venom induced, and MC disorders. It has been thought that intrinsic abnormalities in MCs, including the presence of the activating KIT D816V mutation in mastocytosis or of genetic trait, hereditary alpha-tryptasemia, may influence susceptibility to severe anaphylaxis.


This special issue aims to collect papers that evaluate the potential mechanisms leading to severe MC activation, as well as the differential diagnosis of range of symptoms and conditions attributable to systemic MC mediator release including anaphylaxis, mastocytosis and MC activation syndromes.


Thus, we would like to invite you to contribute, either in the form of original research articles, reviews or shorter perspective articles on all aspects related to the theme of Anaphylaxis and Mast Cell Disorders.
